Phos-FLAG
Phos-FLAG is a phosphine probe containing the Flag peptide. Phos-FLAG undergoes Staudinger ligation with azido groups metabolically incorporated into cell surface glycans and organ glycoproteins, enabling quantitative analysis of the presence of cell surface azides. Phos-FLAG detects azide-labeled glycans in isolated cells and organ lysates.

Phos-FLAG (16 μM; i.p.; single dose; 24 h after final Ac4ManNAz injection) successfully mediates Staudinger ligation with metabolically introduced azide groups on mouse splenocytes in vivo, generating detectable cell-surface Flag epitopes with no observed toxicity[1].

Detected discrete labelled glycoprotein bands in heart, kidney, and liver lysates from Ac4ManNAz-treated mice, but not in brain or thymus homogenates.
Significantly reduced MFI after Phos-Flag labelling in sialidase-treated splenocytes from Ac4ManNAz-treated mice, confirming azide label presence on sialic acids.

Observed no adverse physiological effects in mice after administration.
