Lignin Staining Solution (Pianeze Method)
Lignin Staining Solution (Pianeze Method) is a multichromatic staining solution for plant histology composed of malachite green, Martius yellow and acid fuchsin. Lignin Staining Solution (Pianeze Method) enables lignified secondary walls to appear bright green, non-lignified secondary walls to appear red, and tracheid pit membranes to appear pale red after long-time immersion. Lignin Staining Solution (Pianeze Method) can be used to distinguish lignified cell walls from non-lignified structures and observe the morphology of pit membranes in plant tissue sections such as fern rhizomes. Lignin Staining Solution (Pianeze Method) can be applied to relevant studies on flax wilt and root rot.

Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
Staining Protocol
1.Sectioning & Staining: Prepare thin sections (10-25 µm) of plant material from 70% ethanol and stain in Pianese III-b solution for 18 to 24 hours at room temperature.
2.Differentiation & Clearing: Differentiate with acidified picric acid in 95% ethanol until the wash is pale yellow/light green, then dehydrate through ethanol, clear in xylene, and mount in a permanent medium.
3.Results Interpretation
(1) Lignified/Secondary Walls: Appear in distinct yellowish-green to deep green shades.
(2) Primary Cell Walls & Cytoplasm: Delineated sharply in red/pink hues.
The Lignin Staining Solution (Pianeze Method, specifically Pianeze III b) stains suberin and lignin identically in flax root cortex sections, with no differential staining observed between resistant, partially resistant, and susceptible flax varieties[1].
Lignin Staining Solution (Pianeze Method) differentially stains lignified and unlignified cell wall components in Helminthostachys zeylanica rhizome and root, revealing spindle-shaped pit tori, and stains swollen pit membranes yellow in discolored Botrychium obliquum rhizome tracheids[2].
