PEG2000-DMPE is a polyethylene glycol-lipid conjugate combined with phosphatidylethanolamine. PEG2000-DMPE undergoes a conformational transition from mushroom-like to brush-like at a concentration of 7 mol%, thereby exerting multiple functions such as inhibitor, stabilizer and surface defect modifier. PEG2000-DMPE can form a steric hindrance barrier on the vesicle surface to block calcium ion-induced liposome fusion, and dynamically regulate the fusion process through spontaneous transfer rate. PEG2000-DMPE can delay the thinning of foam thin liquid films and smoothly seal surface defects of solid support layers. PEG2000-DMPE can be used in the design and related research of stealth liposomes and micelles.

Biological Activity
PEG2000-DMPE (0.5-2 mole %; 5 min) inhibits calcium-induced fusion of DOPE:POPS LUVs in a dose-dependent manner, with 2 mole % incorporation completely blocking fusion[1].
PEG2000-DMPE (2 mole %; 60 min) has its fusion-blocking effect reversed by addition of POPC sink liposomes, which restores the fusogenic activity of DOPE:POPS LUVs, allowing calcium-induced fusion to proceed after a short lag phase[1].
PEG2000-DMPE (2-10 mole %; 60 min) increases the lag phase before recovery of fusogenic activity in DOPE:POPS LUVs with higher initial incorporation, with 10 mole % preventing fusion recovery entirely even with a POPC sink[1].
PEG2000-DMPE (≤10 mol%, 20 mol%, 35 mol%, 80 mol%) induces conversion of DMPC liposomes to micelles, with complete transition occurring at 35 mol% PEG2000-DMPE in aqueous dispersions at 37 °C, pH 6.8-7.0, and 0.5 M NaCl[2].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (0.93 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (0.93 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
