Southern Blot is a molecular biology technique used to detect DNA sequences. The method includes DNA electrophoresis, transfer to membrane, DNA fixation, DNA probe hybridization and detection. Southern Blot is often used to determine the presence, size, quantity, and structure of target genes. Southern blot is a DNA hybridization assay used to detect a defined DNA sequence within restriction-digested or otherwise fragmented genomic DNA.
