RL560
RL560 is a red-emitting RNA fluorescent dye used for RNA-selective labeling and live-cell RNA imaging. RL560 achieves RNA-selective fluorescence turn-on via covalent acylation between its acylimidazole reactive group and the 2′-OH of RNA ribose, as well as fluorescence signal enhancement through restriction of TICT molecular motion. RL560 is applicable for RNA imaging and multicolor RNA detection, with detection wavelengths of Ex/Em = 560/639 nm.

Operating Instructions

1. Stock Solution Preparation
1.1 Solvent: Anhydrous dimethyl sulfoxide (DMSO).
1.2 Recommended concentration: 1 M.
2. Working Solution Preparation
2.1 Diluent: Cell culture medium.
2.2 Working concentration: 20 μM for cell imaging.
2.3 Notes: Adjust the working solution concentration as needed; prepare and use immediately.
3. Staining Procedures
3.1 Sample types:
3.1.1 In vitro RNA samples: Short single-stranded RNA (ssRNA) oligonucleotides, RNA molecular weight markers, in vitro transcribed RNA.
3.1.2 Cell samples: Adherent HeLa cells.
3.2 Incubation conditions:
3.2.1 Cell imaging: Incubate live HeLa cells with 20 μM RL560 for 30 min without washing.
3.3 Washing step: No washing is required for live cell imaging with RL560.
4. Control Setup
4.1 Negative control: Unreacted carboxylic acid precursor.
5. Detection and Analysis
5.1 Instrument: Laser confocal microscope.
5.1.1 Excitation/emission wavelengths: The excitation wavelength of RNA-labeled RL560 is 560 nm, and the emission wavelength is 639 nm.
5.2 Result analysis:
5.2.1 RL560 produces red fluorescence after covalent binding to RNA, which can be used for live-cell RNA imaging and multicolor RNA labeling.
