TRRAP

TRRAP (transformation/transcription domain-associated protein) is a large adaptor protein within multiple histone acetyltransferase (HAT) complexes, including SAGA/STAGA, PCAF, and Tip60/NuA4, where it coordinates chromatin remodeling and transcriptional activation through histone acetylation-dependent mechanisms[1][2]. Mechanistically, TRRAP functions as a scaffold for transcription factors such as MYC and E2F, promoting recruitment of HAT activities to target promoters and regulating expression programs involved in cell-cycle progression, proliferation, and DNA replication[3][4]. TRRAP also participates in DNA damage responses through its association with the MRN repair complex, linking chromatin modification to double-strand break repair and genome stability pathways[5]. In cellular and murine knockout models, loss of Trrap disrupts mitotic progression, alters expression of genes controlling checkpoint signaling and metabolism, and causes broad defects in histone H3 and H4 acetylation, highlighting its essential role in transcriptional regulation across multiple biological processes[2]. Compared with catalytically active phosphatidylinositol 3-kinase-related kinase family members, TRRAP lacks demonstrated kinase activity and primarily functions as a regulatory cofactor that recruits and stabilizes chromatin-modifying complexes at specific genomic loci[3]. In disease settings, aberrant TRRAP-dependent transcriptional programs contribute to MYC-driven oncogenesis, while pathogenic TRRAP variants have also been associated with neurodevelopmental disorders, supporting its relevance across cancer and developmental biology research[3][6].
