AM89 is a PGC-1α1 stabilizer. AM89 activates the expression of PGC-1α1 target genes and induces the upregulation of Ucp1 gene expression. AM89 is applicable for the research of obesity and related metabolic disorders[1].

In Vitro

AM89 (10 μM; 8 h) increases EGFP-PGC-1α1 fusion protein levels in stable HEK 293-EGFPmPGC-1α1 cells, with an EGFP intensity of 143.3%[1].
AM89 (10 μM; 8 h) induces Ucp1 gene expression 5-fold and modestly increases other PGC-1α1 target gene expression in fully differentiated mouse brown adipocytes[1].
AM89 (10 μM; 8 h) stabilizes endogenous PGC-1α1 protein in fully differentiated mouse brown adipocytes[1].

Real Time qPCR[1]

Cell Line: fully differentiated immortalized mouse brown adipocytes

Incubation Time: 8 h
Result: Induced a 5-fold increase in Ucp1 gene expression relative to DMSO-treated controls.
Induced small increases in the expression of other PGC-1α1 target genes including Tra2, Fsp27, Cytc, Tfam, Cpt1b, Esrra, Glut4, Prdm16, Pparg, and Sod1, though these increases were less pronounced than the Ucp1 induction.
