BTA-EG6 is a brain-penetrant aggregated amyloid-β () peptide binder with a Kd of 290 nM for Aβ1-42. BTA-EG6 binds to aggregated Aβ and forms protein-resistive coatings that block interactions between Aβ and catalase. BTA-EG6 is a brain-penetrant aggregated amyloid-β () peptide binder with a Kd of 290 nM for Aβ1-42. BTA-EG6 binds to aggregated Aβ and forms protein-resistive coatings that block interactions between Aβ and catalase. BTA-EG6 protects neuroblastoma cells from Aβ-induced toxicity and oxidative stress, and inhibits Aβ-induced increases in cellular hydrogen peroxide (H2O2) levels. BTA-EG6 can be used for the research of alzheimer’s disease (AD)[1].

In Vitro

BTA-EG6 (0-500 μM; 24 h) is toxic to differentiated SH-SY5Y human neuroblastoma cells with an IC50 of 69 ± 5 μM after 24 h of treatment[1].
BTA-EG6 (overnight) binds to aggregated amyloid-β 1-42 peptides with a Kd of 290 ± 60 nM following overnight incubation[1].
BTA-EG6 (5-20 μM; 24 h) dose-dependently protects differentiated SH-SY5Y human neuroblastoma cells from 25 μM aggregated amyloid-β 1-42-induced toxicity[1].
BTA-EG6 (1-40 μM; 24 h) dose-dependently reduces 25 μM aggregated amyloid-β 1-42-induced hydrogen peroxide (H2O2) levels in differentiated SH-SY5Y human neuroblastoma cells, normalizing levels at 40 μM after 24 h of treatment[1].
BTA-EG6 (40 μM; 12 h) inhibits the interaction of catalase with immobilized aggregated amyloid-β 1-42 peptides in SH-SY5Y cells[1].

Cell Proliferation Assay[1]

Cell Line: Differentiated SH-SY5Y human neuroblastoma cells exposed to aggregated Aβ 1-42 peptides
Concentration: 5, 10, 20 μM (in presence of 25 μM aggregated Aβ 1-42)
Incubation Time: 24 h
Result: Increased cell viability in a dose-dependent manner, over 80% at 20 μM, which was statistically significant compared to cells treated with Aβ alone.

Immunofluorescence[1]

Cell Line: SH-SY5Y cells
Concentration: 40 μM
Incubation Time: 12 h
Result: Reduced fluorescence intensity of amyloid-β 1-42.
In Vivo

BTA-EG6 crosses the blood-brain barrier, decreases in vivo Aβ levels, and improves memory and learning in a mouse model of Alzheimer's disease[1].
