NOTA-FZCD-3 is a NOTA (HY-134418)-labeled FZCD-3 polypeptide precursor that targets cathepsin D (CTSD) with a KD of 0.65 μM. NOTA-FZCD-3 binds specifically to the active site of CTSD, exhibits rapid in vivo clearance properties, and remains stable in blood for more than 2 h. NOTA-FZCD-3 can be used in studies monitoring CTSD-positive tumors.

[68Ga]Ga-NOTA-FZCD-3 (0.5-2 h; 37°C) remains stable for at least 2 hours at 37°C in normal saline and human serum, with no decomposition detected[1].
[68Ga]Ga-NOTA-FZCD-3 (0.5-2 h) is taken up by human breast cancer cell lines MCF-7, MDA-MB-468, MDA-MB-415 and MDA-MB-175VII in a time-dependent manner. The uptake level is directly correlated with endogenous CTSD expression, and a peak uptake rate of 1.45% is observed in MDA-MB-175VII cells at 1 h[1].

In Vivo
[68Ga]Ga-NOTA-FZCD-3 (2.5-7.4 MBq; intravenous injection; single administration) exhibits the highest tumor uptake rate and contrast in the MDA-MB-175VII breast cancer xenograft model, with a peak tumor-to-non-tissue ratio of 7.65 at 1 h post-injection[1].
[68Ga]Ga-NOTA-FZCD-3 (3.7-7.4 MBq; intravenous injection; single dose) achieves specific tumor uptake in A549 lung cancer xenografts, with an initial uptake of 2.47% ID/g at 0.5 h post-injection[1].
The uptake of [68Ga]Ga-NOTA-FZCD-3 (2.5-3.7 MBq; intravenous injection; single administration) in MDA-MB-175VII tumor xenografts is directly correlated with endogenous CTSD expression levels[1].
[68Ga]Ga-NOTA-FZCD-3 (intravenous injection; single dose) exhibits favorable biosafety in tumor-bearing BALB/c nude mice, with no organ toxicity detected 7 days post-injection[1].

Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (1.99 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (1.99 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
