Xanthopurpurin is an orally active anthraquinone glycoside. Xanthopurpurin can be isolated from the rhizome of Rubia akane. Xanthopurpurin has antiviral effects against rotavirus and HIV. Xanthopurpurin has a strong inhibitory effect on collagen-induced platelet aggregation. Xanthopurpurin prevents peanut allergy.

In Vitro
Xanthopurpurin (2.5-40 μg/mL) inhibits IgE production in a dose-dependent manner in human myeloma U266 cells[1].
Xanthopurpurin (15 μg/mL) shows 42% inhibition of HIV in CEM-GFP cells infected with HIV-1[2].
Xanthopurpurin is very efficient against rotavirus in MA104 cells by enhancing virus-mediated apoptosis and suppressing its proliferation[3].
Xanthopurpurin shows strong inhibition of collagen-induced platelet aggregation in washed rabbit platelets[4].

Animal Model:Six-week-old female C3H/HeJ mice, peanut-allergic murine model[1]

Dosage:200 μg or 400 μg per mouse per day (dissolved in drinking water)

Administration:Orally administered, for 4 weeks

Result:Significantly reduced peanut-specific IgE levels, plasma histamine levels, and protected the mice against peanut-allergic reactions in both early and late treatment experiments.
Increased IL-4 promoter methylation, reduced the number of peripheral and bone marrow IgE+ B cells, and regulated the gene expression of CCND1, DUSP4, SDC1, ETS1, PTPRC, and IL6R related to plasma cell IgE production.

Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 1.25 mg/mL (5.20 mM); Clear solution
This protocol yields a clear solution of ≥ 1.25 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (12.5 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
