PSMA-1-DOTA (1 h) inhibits ³H-ZJ24 binding to PC3pip cells with an IC₅₀ of 1.83 nM, showing approximately fourfold greater affinity for PSMA than PSMA-I&T and PSMA-11^[1].

In Vivo

PSMA-1-DOTA (1.4 mg/kg; i.v.; single dose) is well tolerated in NMRI mice with minor histopathological changes in lymphoid tissues^[1].
[¹⁷⁷Lu]Lu-PSMA-1-DOTA (111 MBq total, 4 doses of 27.75 MBq over 2 days; 111 MBq single dose; i.v.) effectively inhibits tumor growth in PC3pip xenograft mice for 24 days^[1].
[⁶⁸Ga]Ga-PSMA-1-DOTA (7.4 MBq; i.v.) targets PC3pip tumors with minimal non-target uptake in kidneys and salivary glands in xenograft mice^[1].
