After covalently binding to CRBN, CRBN ligand-186 (compound B1) can act as a molecular glue to alter the surface conformation of CRBN and enhance its interaction with target proteins; alternatively, it can be connected via a linker to a target protein ligand to form heterobifunctional degraders (PROTACs), which recruit specific target proteins to the E3 ligase complex, thereby triggering ubiquitination and proteasomal degradation of the target proteins[1].
