Cy5 DBCO chloride is an azide reaction probe and the addition of DBCO molecules allows the imaging of azide-labelled biomolecules by a copper-free "Click Chemistry" reaction[1]. Cy5 DBCO (chloride) is a click chemistry reagent, it contains a DBCO group that can undergo strain-promoted alkyne-azide cycloaddition (SPAAC) with molecules containing Azide groups.

In Vitro

Cy5 DBCO chloride (2-50 μM) labels cells in a dose-dependent manner, and the amount of DBCO-Cy5 bound to the cell surface increases with increasing DBCO-Cy5 concentration[1].
Guide (The following is a recommended protocol for reference only and should be adjusted according to your specific experimental needs).
1. Seed the cells in a culture dish and culture for 3 days, then wash the cells twice with DPBS (pH 7.4).
2. Add Cy5 DBCO chloride at a final concentration of 20 μM and incubate at 37℃ for 1 h.
3. After incubation, wash the cells with DPBS (pH 7.4), then fix them with formaldehyde-glutaraldehyde combined fixative at room temperature for 15 min.
4. After fixation, wash the cells twice with DPBS (pH 7.4), and analyze the samples using a flow cytometer, fluorescence microscope, or fluorescence microplate reader.
