Cy7 tyramide, a red fluorescent dye, is utilized as reporter fluorescent substrate for horseradish peroxidase (HRP)-catalyzed deposition that is signal amplification technique in immunoassay and in situ hybridization of nucleic acids.

Biological Activity
Guide (The following is our recommended solution. This solution is merely a guideline and should be modified according to your specific needs.)
The cells were fixed in a 1x PBS solution containing 3.7% paraformaldehyde at room temperature for 20 minutes, and then washed twice with 1x PBS.
2) Incubate in 0.1% Triton X-100 at room temperature for 1 to 5 minutes, then wash twice with 1x PBS.
3) Incubate in a 1x PBS solution containing 3% H2O2 for 10 minutes to block endogenous peroxidase activity, and then wash twice with 1x PBS.
4) Incubate at 4°C for 30 minutes in a 1x PBS solution containing 1% BSA.
5) Add the primary antibody working solution, incubate at room temperature for 1 hour or at 4°C overnight, and then rinse three times with 1x PBS, each time for 5 minutes.
6) Add 100 μL of the secondary antibody-HRP working solution, incubate at room temperature for 1 hour, then wash 3 times with 1x PBS, each time for 5 minutes.
7) Add 100 μL of Cy7 tyramide working solution, incubate at room temperature for 5-10 minutes, then rinse 3 times with 1x PBS, each rinse for 5 minutes.
8) Fluorescence microscopy detection.
Note: It is recommended to determine the optimal concentration of Cy7 tyramide working solution based on the specific circumstances.
