MAP3K6/ASK2

MAP3K6 (ASK2) is the closest paralog of ASK1 within the STE11 group and functions largely as its heterodimerization partner, forming ASK1-ASK2 complexes whose activity and stress responsiveness differ from those of ASK1 homodimers. ASK2 can phosphorylate MKK3/6 and MKK4/7, but its independent signaling output is weaker, and evidence points to roles in modulating oxidative-stress, osmotic-stress and apoptotic responses in epithelial tissues, where its expression is prominent. Reduced ASK2 levels have been reported in skin and other epithelial cancers, suggesting a growth-suppressive function. Recombinant ASK2 is used to reconstitute ASK-family heterocomplexes and dissect their differential regulation.
