Biological Activity
DMG-PEG5000-CTP is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against brain endothelial cell receptors/CNS-specific transport mechanisms.
Composition and role of each component:
1. DMG (dimyristoylglycerol): Lipid anchoring group-a diacylglycerol with two C14 chains, providing moderate membrane anchorage; its short-chain, high-fluidity characteristics enable faster dissociation of PEG-lipids from nanoparticle surfaces, facilitating endosomal escape and cellular uptake.
2. PEG5000 (polyethylene glycol, MW 5000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. CTP: Targeting ligand-can be reversibly linked (e.g., via enolase, thiol groups) to a carrier (e.g., miRNA), thereby enabling specific delivery to cardiomyocytes rather than all other organs.
