It can be used as a positive reference for IVT (in vitro transcription) or LNP delivery systems.

In Vitro

eGFP mRNA (4 µg per 2 × 106 cells; 30 min-5 d) electroporated into cord blood CD34+ cell-derived dendritic cells (under 1.75 kV/cm, 250 µs, single pulse conditions) achieves >90% transfection efficiency, induces sustained EGFP expression for up to 5 d, alters surface marker expression, and does not change allogeneic antigen-presenting capacity, with 97.7% cell viability post-electroporation[1].
eGFP mRNA (4 µg per 2 × 106 cells; 24 h) electroporated into peripheral blood adherent cell-derived dendritic cells (under 1.75 kV/cm, 250 µs, single pulse conditions) increases CD80, CD83, CD86, and HLA-DR surface expression without altering CD1a levels when analyzed 24 h post-transfection[1].
eGFP mRNA (4 µg per 2 × 106 cells; 24 h) electroporated into peripheral blood CD14+ cell-derived dendritic cells (under 1.75 kV/cm, 250 µs, single pulse conditions) slightly reduces CD1a expression, increases CD80, CD83, CD86, and HLA-DR expression, and does not change allogeneic antigen-presenting capacity when analyzed 24 h post-transfection[1].
