Elotuzumab (HuLuc 63) is an IgG1 monoclonal antibody targeting the SLAMF7 receptor. Elotuzumab exerts antitumor activity by activating natural killer cells and inducing antibody-dependent cell-mediated cytotoxicity. Elotuzumab can be combined with Lenalidomide (HY-A0003), Dexamethasone (HY-14648), etc., for the research of tumors such as multiple myeloma.

Elotuzumab (0.1-10 μg/mL; 4-20 h) upregulates the expression of CD107a, CD69 and CD54 on NK cells, and promotes the secretion of IFN-γ and TNF-α in a co-culture system of human PBMC and OPM-2 cells[2].

Animal Model:Female BALB/c mice treated A20-hSLAMF7 cells or EG7-hSLAMF7 cells[2]

Dosage:10 mg/kg (Elotuzumab in the form of murine IgG2a subtype)

Administration:Intraperitoneal injection; 3 times; once every 3-4 days

Result:Inhibited tumor growth, increased tumor-infiltrating NK cells and CD8+ T cells, and the expression of activation markers on these cells.
Promoted tumor-infiltrating NK and CD81 T-cell activation, as well as increased intratumoral cytokine and chemokine release by combination with anti-PD-1 antibody.

Immobilized Human CS1 can bind Elotuzumab. The EC50 for this effect is 20.87 ng/mL. -
Flow cytometric analysis of 1X106 MM.1S cells with Elotuzumab (HY-P9965, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

Mammalian live/dead viability and cytotoxicity staining
Live/dead viability and cytotoxicity staining assays are based on the simultaneous detection of intracellular esterase activity in metabolically active (viable) cells and membrane integrity loss in non-viable cells. In commonly used dual-staining approaches, membrane-permeant fluorogenic substrates are converted by intracellular esterases into fluorescent products in live cells, while impermeant DNA-binding dyes selectively enter cells with compromised plasma membranes and label nucleic acids in dead or dying cells, enabling discrimination between viable and non-viable populations by fluorescence microscopy or flow cytometry.

Cell Viability Determination by MTT Colorimetric Assay
The following protocol uses the MTT colorimetric assay as a classic literature-established method for assessing cell viability/metabolic activity in cultured mammalian cells. MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] is reduced by metabolically active cells to a colored formazan product; the amount of formazan is quantified spectrophotometrically and provides an indirect measure of metabolically active viable cells. Importantly, MTT reduction reflects cellular oxidoreductase/metabolic activity rather than an absolute direct count of living cells, so changes in cellular metabolism can alter the signal independently of cell number.
