F-P564
F-P564 is a FITC (HY-66019)-labeled HIF-1α-derived peptide substrate used to monitor PHD2 prolyl hydroxylase activity. F-P564 undergoes PHD2-mediated hydroxylation of Pro-564 to generate a hydroxyproline-containing peptide, which binds the VBC complex, increasing mass and enhancing fluorescence polarization in the assay. The unhydroxylated form of F-P564 shows no change in fluorescence polarization upon addition of VBC.

In Vitro
F-P564 (5 µM; 2 h) is hydroxylated by PHD2 in cell-free MALDI-TOF assays, producing a 2791-to-2807 Da mass shift that is modulated by CQ and Zn2+[1].
F-P564 (1 μM; 30 min) is hydroxylated by PHD2, producing a mass shift from 2793 to 2808 and enabling VBC binding in the fluorescence polarization assay[3].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
