Fazadinium bromide (AH 8165 bromide) is a species-specific neuromuscular blocking agent that primarily targets the acetylcholine receptor (AChR) at the skeletal neuromuscular junction, blocking the receptor agonist binding site in a competitive and reversible manner. Fazadinium bromide induces sustained tetanic fade, and tetanic contractions are more sensitive than single twitches. Fazadinium bromide can be used in anesthesia and research related to neuromuscular transmission[1].

In Vivo

Fazadinium bromide (0.125-4.0 mg/kg; i.v.) produces complete neuromuscular blockade in anaesthetized cats at 2.0 mg/kg i.v. with a 1-minute onset and 15-minute mean recovery time, while inducing vagal blockade at lower doses and minimal sympathetic blockade at supramaximal doses[1].
Fazadinium bromide (0.0625-4.0 mg/kg; i.v.) is four times more potent in anaesthetized rhesus monkeys than in cats, producing complete neuromuscular blockade at 0.5 mg/kg i.v. with a 1.5-5 minute onset and 36-minute mean recovery time, while inducing vagal blockade at near-paralyzing doses and sympathetic blockade at supramaximal doses[1].
