QS-21-Api, an immunostimulatory saponin, could be used as a potent vaccine adjuvant. QS-21-Api stimulates Th2 humoral and Th1 cell-mediated immune responses through action on antigen presenting cells (APCs) and T cells. QS-21-Api can activate the NLRP3 inflammasome with subsequent release of caspase-1 dependent cytokines, IL-1β and IL-18.

PI staining assay. SMMC cells were incubated with different concentrations (1, 2.5, 5, 7.5, and 10 μM) of QS-21-Api (QS-21) for 24 h. Broken cells were stained with PI, and images were taken by fluorescence microscopy. The proportion of PI-positive cells was quantified by CellProfiler. Treatment of SMMC cells with different concentrations of QS-21 resulted in less than 15% PI-positive cells.

Cytoplasmic Ca2+ assay. HeLa cells were incubated with different concentrations of QS-21-Api (QS-21) for 24 h, followed by staining with Fluo-4 AM. Ca2+ was detected by fluorescence microscopy or flow cytometry. Cytosolic Ca2+ concentrations significantly increased in a concentration-dependent manner after treatment with 2.5–5 μM QS-21.

HeLa cells were treated with different concentrations (2.5, 7.5, and 15 μM) of QS-21-Api (QS-21), MAP30 (10 nM), or QS-21 (2.5 μM) + MAP30 (10 nM) for 6 h. The LC3-I and LC3-II levels were detected by Western blot assay. QS-21 could induce the conversion of LC3-I to LC3-II in a dose-dependent manner, which was not observed upon MAP30 treatment, indicating that this autophagy was induced by QS-21.

Colocalization detection of lysosomes and autophagosomes. HeLa cells were transfected with the GFP-LC3 plasmid for 24 h, followed by treatment with QS-21-Api (QS-21) (2.5, 7.5 μM), MAP30 (10 nM), or QS-21(2.5 μM) + MAP30 (10 nM) for 6 h. Lysosomes were labeled by the rabbit anti-LAMP1 antibody. Images were captured by CLSM. After treatment with QS-21 or QS-21+MAP30, noticeable autophagosomes appeared in HeLa cells transfected with the LC3 gene and significantly colocalized with lysosomes.

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (1.26 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (1.26 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
For the following dissolution methods, please prepare the working solution directly:
It is recommended to prepare fresh solutions and use them promptly within a short period of time.
