Description

GFP11 is a protein fusion tag consisting of 16 amino acids. When GFP11 and GFP1-10 are sufficiently close in physical space, they undergo spontaneous, irreversible complementation to reassemble into an intact, functional GFP protein that emits green fluorescence. GFP11 can be used to form a split GFP system for high-throughput biotechnology and flow cytometry applications[1][2].

In Vitro

GFP11 can deliver proteins to the outer membrane of Gram-negative bacteria, and enables them to emit green fluorescence via complementation with exogenously added GFP1-10. Due to its low molecular weight, GFP11 does not significantly affect protein solubility and expression, nor does it alter the catalytic activity of the enzyme CsBglA[1].
With its extremely short DNA sequence, GFP11 can be efficiently and inexpensively integrated into endogenous gene loci via CRISPR/Cas9-mediated homology-directed repair, thereby enabling fluorescent labeling of endogenous proteins and providing a technical basis for constructing protein localization libraries[2].
