IB15C is an ILT3 (LILRB4) inhibitor with a human Kd of 0.92 μM. IB15C binds to a specific ILT3 pocket, disrupts ApoE interaction, and interferes with downstream inhibitory signaling pathways. IB15C binds to recombinant human ILT3 extracellular domain with submicromolar affinity, showing an apparent Kd of 0.92 μM[1].
IB15C inhibits the interaction between recombinant human ILT3 extracellular domain and ApoE in an ELISA-based assay with an IC50 of 0.48 μM[1].
IB15C inhibits the interaction between recombinant human ILT3 extracellular domain and ApoE in a BLI-based real-time assay with an IC50 of 0.64 μM[1].
IB15C (1-10 μM) reverses ILT3-dependent inhibitory signaling, reduces inflammatory activation, and restores Aβ uptake in human iPSC-derived microglia under Aβ42- and ApoE-driven conditions, with no detectable cytotoxicity at concentrations up to 10 μM[1].
