(S)-DMP2
(S)-DMP2 is a benzoxaborole-derived photoaffinity probe that efficiently enriches eukaryotic translation initiation factor 4E (eIF4E) with stereoselectivity. (S)-DMP2 acts as a cap-binding competitive inhibitor that competes with the 7-methylguanosine cap of mRNA for binding at the cap-binding pocket of eIF4E, covalently modifies residues within this pocket, and impairs the cap-binding ability of eIF4E. (S)-DMP2 is applicable to research on eIF4E-related mechanisms and diseases.

(S)-DMP2 selectively enriches eIF4E in HEK293T cells[1].
(S)-DMP2 (5-100 μM) labels eIF4E in HEK293T cells in a dose-dependent manner. This labeling disrupts the epitope recognized by mouse anti-eIF4E antibody, but does not affect the epitope recognized by rabbit anti-eIF4E antibody[1].
(S)-DMP2 specifically binds to the cap-binding pocket of eIF4E and labels recombinant eIF4E in vitro, and this labeling process can be competitively blocked by m7GTPG and Taunton 12; in cellular assays, pretreatment with Taunton 12 inhibits the labeling of (S)-DMP2 and restores the binding of eIF4E antibody[1].
(S)-DMP2 tag impairs the cap-binding activity of eIF4E in HEK293T cells, as evidenced by the reduced binding of eIF4E to m7GTP-modified agarose microspheres[1].
(S)-DMP2 covalently modifies the Met101 and Glu103 residues within the cap-binding pocket of recombinantly expressed eIF4E[1].
