  3. Penicillinase

Penicillinase is an enzyme capable of hydrolyzing penicillin. Penicillinase converts penicillin into penicilloic acid, which has no antigenicity, and destroys the antibacterial, antigenic and epileptogenic properties of penicillin. Penicillinase shortens the duration of penicillin-induced seizures and neutralizes the ability of penicillin to form epileptogenic foci. Penicillinase can be used in research related to penicillin hypersensitivity and penicillin-induced encephalopathy.

Description

Penicillinase is an enzyme capable of hydrolyzing penicillin. Penicillinase converts penicillin into penicilloic acid, which has no antigenicity, and destroys the antibacterial, antigenic and epileptogenic properties of penicillin. Penicillinase shortens the duration of penicillin-induced seizures and neutralizes the ability of penicillin to form epileptogenic foci. Penicillinase can be used in research related to penicillin hypersensitivity and penicillin-induced encephalopathy[1][2].

In Vitro

Penicillinase destroys penicillin in vitro[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

In Vivo

Penicillinase (800,000 units; intravenous or intramuscular injection) completely eliminates penicillin-induced generalized epileptiform electroencephalographic and behavioral seizure activity in adult cats within 3 hours after the first injection of penicillin, and significantly shortens the seizure duration that exceeds 6 hours without treatment[2].
Topical administration of penicillinase prevents or eliminates penicillin-induced focal epileptogenic foci in acutely prepared adult cats without altering normal cortical electroencephalographic activity, but it has no effect on strychnine-induced epilepsy[2].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: unspecified strain (adult, epilepsy induced by intramuscular injection of 325,000 units/kg aqueous Na penicillin G)[2]
Dosage: 800,000 units
Administration: i.v., over 2 minutes; i.m., single dose
Result: Reduced frequency of penicillin-induced bilaterally synchronous 4- to 5-per-second spike-wave bursts and associated behavioral twitches, with complete disappearance by 3 hours after initial penicillin injection.
Eliminated penicillin-induced seizure activity without tachycardia or other side effects when administered i.m.
Shortened the duration of EEG abnormalities from at least six hours (untreated) to 3 hours.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
