In Vitro

Guidelines (The following are our recommended protocols. This protocol is for reference only and should be modified according to your specific needs.)
General Protocol
1.1 Preparation of PKH 26 Working Solution
Dilute with serum-free cell culture medium or PBS. Adjust the stock solution as needed. Note that if the solvent is DMSO, the cytotoxicity of DMSO must be considered, and a solvent control should be prepared; if the solvent is pure water, the working solution needs to be filtered and sterilized before adding cells.
Note: Adjust the concentration of PKH 26 working solution as needed.
2. Cell Staining
2.1 Suspension Cells (6-well plate)
a. Centrifuge at 4°C, 1000 g for 3-5 minutes, discard the supernatant, wash twice with PBS for 5 minutes each time. Cell density is 1×10?/mL.
b. Add 1 mL of working solution and incubate at room temperature for 10-45 minutes.
c. Centrifuge at 400 g for 3-4 minutes at 4°C and discard the supernatant.
d. Wash twice with PBS, 5 minutes each time.
e. Resuspend the cells in serum-free cell culture medium or PBS. Observe under a fluorescence microscope or flow cytometry.
2.2 Adherent Cells
a. Culture adherent cells on sterile coverslips.
b. Remove the coverslips from the culture medium and aspirate any excess medium.
c. Add 100 μL of working solution, gently shake to completely cover the cells, and incubate at room temperature for 5-30 minutes.
d. Wash twice with culture medium, 5 minutes each time. Observe under a fluorescence microscope or flow cytometry.

Storage
It is recommended to store the stock solution at -80°C, protected from light, and avoid repeated freeze-thaw cycles.
