Propiolamide is a monooxygenase (MMO) system activator and glutathione peroxidase 4 (GPX4) inhibitor. Propiolamide induces ROS production through interaction with the MMO system. Propiolamide promotes the accumulation of intracellular cytotoxic lipid peroxides and induces ferroptosis. Propiolamide induces programmed cell death via the apoptosis pathway. Propiolamide can be used in cancer research.

Propiolamide (14 mM; 0-30 min) induces microsomal monooxygenase-mediated reactive oxygen species production in CYP3A4/CPR/Cyt c and CYP3A4/NADPH systems[1].
Propiolamide (38 μM) inhibits the activity of glutathione peroxidase 4 derived from bovine red blood cells by 46%[1].
Propiolamide (30-50 μM; 12 h) induces apoptosis in HeLa cells in a concentration-dependent manner in vitro. After 12 hours of treatment, the proportion of total apoptotic cells reaches 7.13% at 30 μM and 10.33% at 50 μM[1].
Propiolamide (50 μM; 12 h) promotes lipid peroxidation in HeLa cells, which is a key marker of ferroptosis[1].
