Prostaglandin D1
Based on 1 Customer Validation
Prostaglandin D1 (PGD1) is a CatSper activator that triggers Ca2+ influx through sperm-specific Ca2+ channels via the prostaglandin binding site and competes with other prostaglandins.

Biological Activity
Description
In Vitro
Prostaglandin D1 (up to 100 μM) activates CatSper in human sperm with very low potency, failing to reach saturation at 100 μM, and shows increased efficacy compared to PGE1, with the response abolished in CATSPER2-deficient sperm[1].
Prostaglandin D1 competes with PGE1 for the same prostaglandin-binding site to activate CatSper in human sperm, with an IC50 of 0.7 μM for suppressing the PGE1-induced Ca2+ signal[1].

Ca2+ Staining Technique
Ca2+ staining is an experimental technique that utilizes specific fluorescent probes (such as Fluo-4 AM, Fura-2, etc.) to qualitatively or quantitatively detect dynamic changes in intracellular Ca2+ concentrations; this is achieved by monitoring the changes in fluorescent signals generated when these probes bind to free intracellular calcium ions. The underlying principle relies primarily on the presence of chelating groups within the probe's molecular structure that possess high affinity for calcium ions.
