ADAM29

ADAM29 is one of the least characterized members of the reproductive ADAM branch, sharing testis-restricted expression with ADAM20. Its domain architecture mirrors that of other membrane ADAMs, including a disintegrin domain poised to mediate adhesion during gamete interaction. No physiological substrate has been assigned, and loss-of-function phenotypes remain to be established, so current knowledge rests largely on sequence and expression analyses. Recombinant ADAM29 therefore serves primarily as a reagent for cataloging sperm-surface components and for probing adhesion events in fertilization and contraceptive-target studies. Its prodomain carries the cysteine switch that keeps the enzyme latent until activation, and the mature protein is predicted to be anchored in the germ-cell membrane by a C-terminal transmembrane region.
