CELSR1
CELSR1, also called Flamingo-1 or EGF-like repeat and discoidin-like receptor 1, is an atypical cadherin that combines an extracellular region of cadherin repeats, EGF-like domains, and laminin G domains with seven transmembrane segments and a GPCR-like tail. It is a core component of the planar cell polarity pathway, acting alongside Frizzled and Vangl proteins to orient cells within the epithelial plane. CELSR1 governs convergent extension and neural tube closure during development, and heterozygous mutations have been found in families with lymphedema and neural tube defects. These roles distinguish CELSR1 from classical cadherins, which lack both the seven-pass topology and direct polarity-pathway coupling.
