PIK3CD

p110δ, encoded by PIK3CD, is the hematopoietic-restricted catalytic subunit of class IA PI3K. It signals downstream of the B-cell receptor, co-receptors and cytokine receptors in leukocytes, producing PIP3 that engages AKT and its effectors to control lymphocyte activation, proliferation and migration. Because of this near-exclusive expression in blood cells, p110δ became a landmark immuno-oncology target: the selective inhibitor idelalisib demonstrated clinical benefit in B-cell malignancies, and further p110δ inhibitors are in development for lymphoma, leukemia and autoimmune disease. Germline gain-of-function mutations cause the immunodeficiency syndrome APDS. Its leukocyte-selective biology additionally positions p110δ in chronic inflammatory and allergic airway responses.
