Rhodamine 101 chloride (Rhodamine 640 chloride) is a fluorescent dye with excitation and emission maxima at 565 nM and 595 nM, respectively. Rhodamine 101 chloride binds to intracellular proteins and emits specific fluorescence. Rhodamine 101 chloride can be used in colon cancer research.

Biological Activity
Guide (The following is our recommended protocol. This protocol is for guidance only and should be modified according to your specific needs).
1 Preparation of stock solution
1) Weigh approximately 3-5 mg of Rhodamine 101 powder into 0.3 mL of DMSO.
2) Add distilled water to the above solution to adjust the final concentration of Rhodamine 101 to 1 mg/mL.
2 Preparation of working solution (for staining ethanol-fixed cells in combination with Mithramycin)
1) Dilute the stock solution with 0.15 M sodium chloride aqueous solution to adjust the final concentration of Rhodamine 101 to 1.0 μg/mL.
3 Specific Staining Steps
1) Prepare ethanol-fixed cell samples: Fix target cells (e.g., CHO cells) with 70% ethanol for at least 18 hours. Centrifuge the cell suspension to remove the ethanol fixative.
2) Resuspend the centrifuged cells in an appropriate amount of the prepared Rhodamine 101-Mithramycin dual-dye working solution.
3) Incubate the cell-dye mixture at room temperature for at least 10 minutes to ensure adequate staining.
4) After incubation, analyze the stained cells using a flow cytometer.
