Description

SDSSD is an osteoblast-targeting peptide. SDSSD selectively binds to periostin on osteoblasts and mediates targeted delivery to these cells. SDSSD can endow calcium phosphate nanocomposites with bone-targeting properties, and these materials exhibit osteointegrative capacity in both in vitro and in vivo models. When conjugated with polyurethane nanomicelles, SDSSD enables targeted delivery of siRNA/microRNA to osteoblasts[1][2].

In Vitro

SDSSD colocalizes with periostin in murine MC3T3-E1 clone 4 osteoblasts and human hFOB 1.19 osteoblasts[2].
SDSSD reduces the subsequent binding of FITC-labeled SDSSD peptide to periostin in mouse MC3T3-E1 clone 4 osteoblasts and human hFOB 1.19 osteoblasts[2].
SDSSD−PU efficiently delivers FAM-labeled siRNA to murine MC3T3-E1 clone 4 osteoblasts and human hFOB 1.19 osteoblasts, and the uptake process depends on periostin binding[2].

In Vivo

SDSSD-modified polyurethane nanomicelles delivering anti-miR-214 (10 mg/kg; tail vein injection; every 1 week; 1 month) reduce osteoblast miR-214 levels by 80% and significantly increase bone mineral density, mineral apposition rate, and bone mass in ovariectomized mice with osteoporosis[2].
SDSSD-modified polyurethane nanomicelles delivering GFP siRNA (10 mg/kg; tail vein injection) selectively reduce GFP mRNA and protein levels in bone tissue and isolated osteoblasts of GFP transgenic mice, demonstrating targeted gene knockdown efficacy[2].
