Humantenine is a highly toxic indole alkaloid from Gelsemium elegans (Gardn. & Champ.) Benth. that binds to RNA m6A modification regulatory proteins (ALKBH5, METTL). Humantenine stably binds via hydrogen bonding and hydrophobic interactions and disrupts the m6A methylation level of target genes, thereby impairing the expression of intestinal epithelial cell tight junction and cytoskeleton-related genes, causing intestinal barrier dysfunction and significant intestinal cytotoxicity. The intraperitoneal injection LD50 values of Humantenine are <1 mg/kg in mice, 1.2 mg/kg in male rats and 1.5 mg/kg in female rats, respectively. Species differences exist in the metabolism of Humantenine in human, porcine, goat and rat liver microsomes, and demethylation, dehydrogenation and oxidation occur in liver microsomes .

Lys-VBOD is a Fluorescent probe for lysosomal viscosity monitoring and live cell imaging. Its detection mechanism depends on viscosity: increased viscosity restricts the twisting of the double bond between the indole and BODIPY fluorophore, as well as the rotation of the single bond between quinoline and BODIPY, which reduces non-radiative decay and leads to enhanced fluorescence intensity in high viscosity environments; it also features a morpholine group that provides lysosome-targeting properties, allowing it to localize to lysosomes for viscosity detection. The probe has excitation/emission wavelengths of Ex/Em = 602/637 nm, and its fluorescence intensity shows a good linear relationship with log-transformed viscosity values. Lys-VBOD is stable across a wide pH range of 5.35 to 10.74, is not affected by macromolecules and proteins like BSA, has a fluorescence lifetime that increases with viscosity, and can visualize real-time lysosomal viscosity changes in live cells under Dexamethasone (HY-14648) stimulation[1].

LysoAIE2 is a Fluorescent probe for lysosomal viscosity detection and live-cell imaging. Its detection mechanism relies on the aggregation-induced emission effect: it exhibits only weak fluorescence in non-viscous media; in viscous environments, restricted intramolecular motion inhibits non-radiative energy dissipation pathways, thereby significantly enhancing fluorescence intensity. It achieves specific targeting of lysosomes through the proton acceptor property of its indole ring structure, while its hydroxyl group endows it with excellent water solubility. This probe is basically unaffected by microenvironmental polarity and pH within the range of pH 4.0 to pH 8.0, which avoids interference from these factors in viscosity measurement. LysoAIE2 has an emission wavelength of 570 nm. It can be used to monitor lysosomal viscosity changes during processes such as Dexamethasone (HY-14648)-induced lysosomal migration and starvation-induced mitophagy in live cells; at concentrations up to 40 μM, cell viability remains above 80%, demonstrating excellent biocompatibility .

L-699333 is a 5-lipoxygenase (5-LO) inhibitor belonging to the thieno[2,3,4-cd]indole class. This compound has a 2-ethoxybutyric acid side chain and is a potent inhibitor of the biosynthesis of 5-HPETE and LTB4 produced from human 5-LO, with ICm values of 22 nM, 7 nM, and 3.8 pM for human neutrophils and whole blood, respectively. L-699333 has shown anti-inflammatory and antiasthmatic effects in a variety of animal models, including rat pleurisy models, antigen-induced wheezing models, and awake macaque and sheep asthma models. Its inhibition of 5-LO is highly selective, with higher ICm values or stronger competitive inhibition in FLAP binding assays compared to inhibition of human 15-LO, porcine 12-LO, and ram epididymal cyclooxygenase. The racemic enantiomer 14g of L-699333 is the most potent enantiomer to date, with inhibitory effects similar to those of the known MK-0591, which has been shown in clinical trials to inhibit the biochemical effects of LTB4 biosynthesis in vitro and LTE4 excretion in urine.
