SNX3-IN-1 is a sorting nexin 3 (SNX3) inhibitor. SNX3-IN-1 reduces SNX3 protein expression and inhibits SNX3-mediated activation of the Wnt/β-catenin signaling pathway. Description

SNX3-IN-1 is a sorting nexin 3 (SNX3) inhibitor. SNX3-IN-1 reduces SNX3 protein expression and inhibits SNX3-mediated activation of the Wnt/β-catenin signaling pathway. SNX3-IN-1 inhibits the proliferation and migration of pulmonary fibrosis-related cells, and decreases the expression of fibrosis markers α-SMA and COL-1. SNX3-IN-1 can be used in research related to pulmonary fibrosis[1].

In Vitro

SNX3-IN-1 (Compound LC4) inhibits the enhanced migratory capacity of fibroblasts overexpressing SNX3[1].
SNX3-IN-1 inhibits SNX3-induced β-catenin accumulation and nuclear localization in cells[1].
SNX3-IN-1 (10 μM) inhibits TGF-β1-induced α-SMA expression in AT2 cells and fibroblasts, thereby reducing fibrotic activation[1].

In Vivo

SNX3-IN-1 (Compound LC4) (10 mg/kg; intratracheal administration; once weekly for 3 consecutive weeks) effectively ameliorates Wnt/β-catenin pathway activation, pulmonary function impairment and fibrosis in a mouse model of pulmonary fibrosis[1].

Animal Model: C57BL/6J Snx3-cTg mice (2 months old, male and female; spontaneous pulmonary fibrosis model); C57BL/6J N-Tg mice (2 months old, male and female; bleomycin-induced pulmonary fibrosis model)[1]
Dosage: 10 mg/kg
Administration: intratracheal; once weekly; 3 weeks
Result: Mitigated Bleomycin (BLM) (HY-108345)-induced pulmonary functional injury, reduced fibrosis areas, lowered β-catenin protein levels, and decreased hydroxyproline concentration in N-Tg mice.
Reversed fibrotic effects associated with SNX3 overexpression in Snx3-cTg mice, including reducing lung injury, fibrosis areas, and β-catenin levels.
Demonstrated therapeutic efficacy equivalent to positive control pirfenidone in inhibiting BLM-induced pulmonary fibrosis.
