Tyromycin A
Tyromycin A is an inhibitor targeting Leu-AP, Cys-AP and CP-A, with an IC50 value of 25-41 μg/mL against porcine Leu-AP and an IC50 value of 60 μg/mL against bovine CP-A. Tyromycin A inhibits the hydrolytic activity of target peptidases and exhibits selective activity against Gram-positive bacteria. Tyromycin A moderately inhibits the infectivity of HCV in human hepatocytes, with an IC50 of 6.6 μM, and its inhibitory activity depends on the two maleic anhydride domains. Tyromycin A can be used in studies related to Gram-positive bacterial infections and hepatitis C virus infections.

Tyromycin A (30 min) inhibits leucine aminopeptidase bound to the surface of HeLa S3 cells with a Ki value of 4×10-5 M; it also inhibits cysteine aminopeptidase with a Ki value of 1.3×10-5 M[1].
Tyromycin A inhibits porcine kidney cytosolic leucine aminopeptidase (IC50 25-31 μg/mL), porcine kidney microsomal leucine aminopeptidase (IC50 27-41 μg/mL), and bovine pancreatic carboxypeptidase A (IC50 60 μg/mL) in cell-free assays[1].
Tyromycin A (30 min) weakly inhibits leucine aminopeptidase activity in HeLa (KB3.1) cells, with an IC50 greater than 150 μg/mL at both 50 μM and 100 μM L-Leu-AMC substrate concentrations[2].
