HaloFlipper 30
HaloFlipper 30 is a fluorescent HaloTag fusion protein probe. HaloFlipper 30 reacts with aspartate residues via a terminal chloroalkane to form an ester bond, achieving covalent attachment and membrane insertion; its ethylene glycol linker allows cell penetration, and genetic engineering of the HaloTag fusion protein enables intracellular targeting and labeling of membranes such as the ER, Golgi, and cytoplasm. HaloFlipper 30 enables mechanosensitive membrane tension imaging through planarization of a twisted push-pull mechanoresponsive group in the ground state.
