WEN05-03
WEN05-03 is a EscN ATPase inhibitor. WEN05-03 blocks the active site of EscN ATPase and competitively inhibits its ATP hydrolysis activity. WEN05-03 completely blocks actin cluster formation, reduces actin pedestal formation, and decreases the toxicity of infected mammalian cells. WEN05-03 can be used in studies related to enteropathogenic E. coli (EPEC) infection.

Biological Activity
WEN05-03 acts as a potent competitive inhibitor of purified recombinant enteropathogenic E. coli EscN ATPase, with a Ki value of 16 μM[1].
WEN05-03 (100 μM; 24 h) exhibits extremely low toxicity to HeLa and BALB/3T3 cells, with only 7% growth inhibition observed in BALB/3T3 cells at the concentration of 100 μM, while no significant toxicity is detected in HeLa cells[1].
WEN05-03 (100 μM; 2 h) completely blocks the formation of actin clusters in HeLa cells infected with enteropathogenic E. coli (EPEC), with levels comparable to those in uninfected mock-treated cells[1].
WEN05-03 competitively inhibits the ATP hydrolysis activity of the EPEC EscN type III secretion system (T3SS) ATPase, reduces the toxicity of EPEC to infected HeLa cells, and exerts no effect on bacterial growth or host cell viability[2].
