WT1 126-134 peptide is a Wilms' tumor oncogene protein (WT1) peptide (RMFPNAPYL). WT1 126-134 peptide is presented by HLA-A0201 and induces cytotoxic CD8 T cells capable of killing WT1+ positive tumor cells. WT1 126-134 can form stable complexes with the H-2Db (mouse) or HLA-A0201 (human) molecules. WT1 126-134 peptide/HLA-A0201 complex has an extremely high affinity (K_d = 0.2 nM) with the humanized monoclonal antibody (IgG1). WT1 126-134 peptide can be used as a vaccine for T cells or as a target for antibodies.

WT1 126-134 peptide (100 μg, emulsified with incomplete Freund’s adjuvant, i.p., once a week for 2 weeks) induces epitope-specific CTL, but fails to produce a significant anti-tumor effect in the TRAMP-C2 model^[1].
WT1 126-134 peptide (100 μg, emulsified with incomplete Freund’s adjuvant, i.p., once a week for 3 weeks) combined with the auxiliary peptide (WT1_35-52) significantly enhances CTL induction and delays tumor growth induced by mWT1-C1498 cells in mice^[3].

Cell Viability Determination by MTT Colorimetric Assay
The following protocol uses the MTT colorimetric assay as a classic literature-established method for assessing cell viability/metabolic activity in cultured mammalian cells. MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] is reduced by metabolically active cells to a colored formazan product; the amount of formazan is quantified spectrophotometrically and provides an indirect measure of metabolically active viable cells. Importantly, MTT reduction reflects cellular oxidoreductase/metabolic activity rather than an absolute direct count of living cells, so changes in cellular metabolism can alter the signal independently of cell number.
