Description

4-Methylumbelliferyl phosphate (4-MUP), an anionic organophosphate, is a acid and alkaline phosphatase fluorogenic substrate. 4-Methylumbelliferyl phosphate is also a nerve agent simulant[1][2][3].

In Vitro

Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
(1) Substrate Preparation: Prepare 4-MUP at a 0.6 mM to 1 mg/mL concentration in a buffer with an alkaline pH (e.g., Tris-NaCl pH 10, or Diethanolamine buffer).
(2) Sample Preparation: Use samples (e.g., lysates, supernatant) diluted in appropriate buffers.
(3) Incubation: Mix 1 part sample with 1-2 parts 4-MUP substrate solution.
(4) Reaction Conditions: Incubate at 37°C or room temperature for 30–60 minutes, protected from light.
(5) Detection: Measure fluorescence at excitation ~ 360 nm and emission ~ 440 nm.
(6) Stopping the Reaction: The reaction can be stopped by adding high pH buffers, such as 0.2 M Glycine/NaOH.
