1. Preparation of working solution
1) Preparation of storage solution
Prepare a 10 mM JAK2 JH2 Tracer stock solution using anhydrous DMSO.
2) Preparation of working solution
Dilute the stored solution with pre-heated serum-free cell culture medium or PBS to prepare a 1.5 pM JAK2 JH2 Tracer working solution.
Note: Please adjust the concentration of JAK2 JH2 Tracer working solution according to the actual situation, and prepare it as needed.
2. Specific staining steps
1) Prepare a series of JAK2 JH2 protein solutions with concentrations ranging from 0 to 4.8 μM in the detection buffer.
2) Add 1.5 pM of JAK2 JH2 Tracer working solution to each well of the microplate.
3) Gradually add the JAK2 JH2 protein solution to the wells containing JAK2 JH2 Tracer, ensuring that the final protein concentration covers the required range.
4) Measure the fluorescence polarization value within 90 minutes.
