Biological Activity
O4I1 (5-20 μM; 1-72 h) potently activates the Oct3/4 promoter in HEK-Oct3/4 cells. After treatment with 5 μM for 72 h, luciferase activity increases to 3-fold of the original level, and the induction shows a sustained, concentration-dependent pattern over time[1].
O4I1 (10-20 μM; 72 h) upregulates the expression of endogenous Oct3/4 mRNA in HEK293 cells, with a 4-fold increase in expression observed after treatment with 20 μM for 72 h[1].
O4I1 (24-96 h) upregulates the protein levels of Oct3/4, Sox2 and Nanog in HEK293 cells in a concentration- and time-dependent manner in vitro. Specifically, Oct3/4 begins to accumulate after treatment with 6 μM for 48 h, and this accumulation persists for at least 96 h under treatment with 10 μM[1].
O4I1 (10 μM; 3-7 days) induces terminally differentiated human fibroblasts to express Oct3/4 protein, including HF cells after 72 h of treatment and HFF cells after 7 days of treatment[1].
O4I1 (10 μM; 72 h) activates the transcriptional activity of Oct3/4 in NCCIT-Oct3/4 cells, increasing luciferase activity by 2.68-fold after treatment at 10 μM for 72 h[1].
