AB054 is a humanized monoclonal antibody targeting factor XII (FXII). In Vitro

AB054 (0-100 μg/mL; 5 min) significantly prolongs the aPTT of human plasma and baboon plasma in a concentration-dependent manner[1].

In Vivo

AB054 (2-5 mg/kg; i.v. bolus; 30 min before experiment) significantly inhibits platelet-dependent thrombosis in baboon membrane oxygenators, and its antithrombotic effect becomes more pronounced when used in combination with Heparin (HY-17567)[1].

Animal Model: Baboons (juvenile male, 9~15kg, with healed surgically implanted chronic arteriovenous shunt device)[1]
Dosage: 5 mg/kg (single agent); 2 mg/kg (plus 5 mg/kg 24h prior, in combination with 20 U/kg heparin)
Administration: i.v. bolus; 30 min before experiment (AB054); 15 min before experiment (heparin)
Result: Reduced platelet deposition rate, terminal platelet deposition, and terminal fibrin content in.
Maintained hemostatic parameters (bleeding time, blood loss, bleeding rate) within normal baboon ranges across all treatment groups.
