AF647 Cadaverine
AF647 Cadaverine is a far-red fluorescent polarity tracer. AF647 Cadaverine possesses a primary amine group that enables covalent labeling of carboxylic acids via carbodiimide activation or covalent labeling of glutamine residues through transamidation, rendering it applicable for bioconjugation, cell imaging, and blood-brain barrier permeability assays (Ex/Em = 651/670 nm).

Biological Activity
Guide (The following is our recommended protocol. This protocol is for guidance only and should be modified according to your specific needs).
1. Preparation: Dissolve your carboxyl-containing biomolecule in a suitable buffer (e.g., PBS or MES, pH 6.0-7.2).
2. Activation: Add 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and sulfo-NHS to activate the carboxyl groups for 1 hour at room temperature.
3. Reaction: Add AF647 Cadaverine dissolved in sodium carbonate buffer (pH ~9).
4. Incubation: React overnight at room temperature in the dark.
5. Purification: Dialyse or use gel filtration (such as Bio-Gel P30) to remove unreacted dye.

Animal Model:C57BL/6 (male and female, 5-7 months of age)[1]
-
Dosage:400 mM
-
Administration:i.v. via retro-orbital vein; single administration per imaging session
-
Result:Detected a 12-fold increase in parenchymal fluorescence in line-scan-induced capillary injury models compared to pre-injection baseline.
Showed no significant increases in parenchymal fluorescence at 5 minutes or 2 days post-single pericyte ablation, indicating no detectable blood-brain barrier leakage.
