In Vivo

α-D-Glucosyl hesperidin (25-50 mg/kg; p.o.; single administration 30 min before torsion reduction) protects Wistar rats against testicular ischemia-reperfusion injury[1].

Animal Model: Wistar (male, 12-14 weeks old, 260-300 g, testicular ischemia-reperfusion injury induced by 2 hours of 720° testicular torsion followed by detorsion)[1]
Dosage: 25 mg/kg; 50 mg/kg
Administration: p.o.; single dose 30 minutes before detorsion
Result: Increased sperm concentration (32.05 ×10⁶/mL), total motility (54.68%), progressive motility (20.53%), kinematic parameters (VAP 23.82 μM/s, VCL 57.78 μM/s, VSL 7.52 μM/s, ALH 4.84 μM/s, BCF 6.95 Hz), membrane functionality (55.48%), and viability (57.29%), while reducing DNA damage (36.15%) and abnormal morphology (34.53%) at 25 mg/kg.
Increased sperm concentration (45.47 ×10⁶/mL), total motility (68.13%), progressive motility (33.45%), kinematic parameters (VAP 28.02 μM/s, VCL 74.89 μM/s, VSL 10.34 μM/s, ALH 5.56 μM/s, BCF 9.06 Hz), membrane functionality (68.47%), and viability (71.87%), while reducing DNA damage (16.93%) and abnormal morphology (23.52%) at 50 mg/kg.
Increased testis weight, epididymis weight, testis/body weight ratio, Johnsen score, seminiferous tubule diameter, Sertoli cell index, repopulation index, mitotic index, Leydig cell nuclear diameter, tubular differentiation index, and spermiogenesis index at 25 mg/kg, while reducing Cosentino score (2.95).
Increased the same reproductive indices more markedly at 50 mg/kg, including testis weight (0.891 g), epididymis weight (0.593 g), testis/body weight ratio (0.34%), Johnsen score (6.64), and seminiferous tubule diameter (195.79 μM), while further reducing Cosentino score (2.13).
Increased total antioxidant capacity (5 mM/mg protein), catalase (0.08 U/mg protein), and superoxide dismutase (18 nmol/mg protein), while reducing glutathione (15 nM/mg protein) and malondialdehyde (500 μM/g protein) at 25 mg/kg.
Increased antioxidant defenses further at 50 mg/kg (TAC 9 mM/mg protein, CAT 0.09 U/mg protein, SOD 22 nM/mg protein), while reducing glutathione (13 nM/mg protein) and malondialdehyde (400 μM/g protein).
Increased Bcl-2 mRNA expression while reducing Bax and caspase-3 expression at both doses, with stronger effects at 50 mg/kg (Bcl-2 0.7-fold; Bax 1.0; caspase-3 1.2).
Increased reproductive performance, including corpora lutea, implants, viable fetuses, pregnancy index, male fertility index, and fertility potential, while reducing preimplantation loss at 25 mg/kg (45.89%) and 50 mg/kg (20.47%), with greater improvement at 50 mg/kg.
