Aminopyridine 2 is an irreversible, selective, and orally active myeloperoxidase (MPO) inhibitor, with an IC50 of 0.16 μM and a Ki of 24 μM against human MPO. Aminopyridine 2 is applicable to research on chronic inflammatory diseases[1].

In Vitro

Aminopyridine 2 (2 h) inhibits the activity of human MPO in plasma, with an apparent IC50 of 1.9 μM[1].

Aminopyridine 2 (2.5-100 mg/kg; p.o.; single administration) dose-dependently inhibits MPO activity in a mouse peritonitis model[1].
Aminopyridine 2 (~50-200 mg/kg; mixed with diet for 14 days) dose-dependently inhibits MPO activity in a mouse peritonitis model[1].

Animal Model: C57BL/6J (male, 8 weeks old)[1]
Dosage: 2.5 mg/kg; 25 mg/kg; 50 mg/kg; 100 mg/kg
Administration: i.g.; single dose
Result: Reduced residual plasma MPO activity by 24% at 2.5 mg/kg, 62% at 25 mg/kg, 74% at 50 mg/kg, and 86% at 100 mg/kg, measured 5 hours post-dose.
Caused dose-dependent inhibition of 3-chlorotyrosine accumulation in peritoneal exudate.
Achieved an apparent in vivo IC50 of 1.4 μM for inhibition of plasma MPO activity.
