LI-3948 is a blood-brain barrier-penetrant IP6K1 inhibitor with an IC50 of 16 nM. LI-3948 reduces locomotor activity and grooming behavior in mice and induces hypothermia. LI-3948 is a moderate P-glycoprotein efflux substrate, with low clearance in rodents and high plasma protein binding. LI-3948 can be used for central nervous system-related research.

In Vitro
LI-3948 (Compound 29c) (30 min) is a potent IP6K1 inhibitor with an IC50 of 16 nM in the ADP-Glo enzymatic assay[1].
LI-3948 (5 μM; 120 min) is a moderate P-gp substrate in MDR1-MDCK cells, with an efflux ratio of 3.8 and an apical-to-basolateral Papp of 16 × 10−6 cm/s[1].

Result:Locomotion was decreased relative to vehicle (t(12) = 2.562) and grooming was decreased (t(12.76) = 2.842).
Body temperature was uniformly and significantly decreased by roughly 3.5 °C measured 24 h after dosing.
Four mice, with similar exposure levels to the other six mice, exhibited locomotor activity similar to vehicle-treated mice.
No statistically significant decrease in IP7 levels in the brain was observed.
