In Vivo

Mavrilimumab (3-100 mg/kg, iv, once a week for at least 11 weeks) inhibits GM-CSF signaling transduction, causes neutral lipids accumulation in alveolar macrophages, the formation of foam cells, cholesterol clefts, and granulomatous inflammation under long-term and high-dosing administration. Mavrilimumab is immunogenic in cynomolgus monkeys, anti-drug antibodies would be produced after multiple doses, which results in accelerated drug clearance and reduced exposure^[2].
