W1507
W1507 is an NH2-TML-based linker-payload conjugate, in which the payload is Exatecan (HY-13631) and the linker is Mal-PEG4-Val-Ala-Ph-2,2-dimethylpropanoic acid (HY-184906). W1507 can be conjugated with Trastuzumab (HY-P9907) to synthesize the ADC Her2-W1507. Her2-W1507 exhibits in vivo anti-tumor activity. W1507 can be used for the research of gastric cancer, ovarian adenocarcinoma and Her2-amplified breast cancer.

W1507 (5 mM; 1 h) is resistant to cathepsin B-mediated cleavage, with no detectable Exatecan (HY-13631) release from the linker-payload in a cell-free in vitro assay[1].
Her2-W1507 (72 h) potently inhibits the proliferation of HER2-positive NCI-N87, SK-OV-3, and SK-BR-3 cells with IC50 values of 48.49 nM, 88.30 nM, and 65.22 nM respectively, while showing minimal activity against HER2-negative MDA-MB-231 cells (IC50 >500 nM) after 72 hours of incubation[1].
