Canocapavir (ZM-H1505R) is an orally active HBV core protein-targeting antiviral agent. Canocapavir binds to the hydrophobic pocket at the dimer-dimer interface of HBV core protein (HBc), stimulating intracellular accumulation of nonfunctional HBV capsids, disrupting viral replication. Canocapavir interferes with the interaction between HBc and HBV large surface protein, resulting in diminished
production of empty virionsis. Canocapavir can be used for the research of hepatitis B virus (HBV) infection.

Canocapavir (135 ng/mL) potently inhibits pan-genotypic HBV replication with a an EC50 of 135 ng/mL[1].
Canocapavir (0.160-5.000 μM; 2 days) potently inhibits HBV replication in HepAD38 Tet-off cells with an EC50 of 0.1185 μM and lacks significant cytotoxicity at concentrations up to 24 μM[2].
Canocapavir (0.02-2 μM; 4 days) dose-dependently inhibits HBV replication in pHBV1.1-transfected HepG2 cells[2].
Canocapavir (3 days) does not affect HBV transcription in HepAD38 Tet-off cells[2].
Canocapavir (0.02-2 μM; 6 days) promotes accumulation of morphologically normal intracellular empty capsids and inhibits HBV pgRNA encapsidation in HepAD38 Tet-off cells[2].
Canocapavir (0.02-2 μM; 2 days) inhibits naked capsid egress and promotes intracellular capsid accumulation in HBc-transfected HepG2 cells[2].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 7.5 mg/mL (13.34 mM); Clear solution
This protocol yields a clear solution of ≥ 7.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (75.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
